Overview

Creatinine is a waste product formed from the normal breakdown of creatine phosphate in muscle tissue. It is produced at a fairly constant rate in the body, depending on muscle mass, and is filtered out of the blood by the kidneys through urine.

The Autozyme Creatinine-SL kit is an advanced, user-friendly diagnostic solution designed for the quantitative determination of creatinine in human serum, plasma, or urine. The kit employs a single liquid reagent format, eliminating the need for multiple reagent preparations and reducing the chance of handling errors. It delivers fast results with excellent linearity and minimal interference, ensuring reliable readings critical for assessing kidney function.

About

The AUTOZYME Creatinine-SL single reagent is intended for the in vitro quantitative diagnostic determination of creatinine in human serum or urine on manual systems.

Key Features:

  • Method: Modified Jaffe’s kinetic method
  • Format: Single liquid reagent – ready to use
  • Sample type: Serum, plasma, or urine
  • Stability: Long shelf-life and onboard stability
  • Benefits: Easy handling, reduced preparation time, and reproducible results
  • Pack Size: 2x50ml and 4x50ml.

This kit is suitable for use on both semi-automatic and fully automated analyzers. Its robustness and compatibility make it a preferred choice for diagnostic laboratories aiming for efficient and cost-effective creatinine testing.

Product Specifications

Creatinine reacts with picric acid under alkaline conditions to form a yellow-red complex. The absorbance of the color produced, measured at a wavelength of 505nm, is directly proportional to creatinine concentration in the sample.

The kit is stable until the expiry date labeled on the bottles when properly stored at 2 – 8°C.

The measuring wavelength is 505nm or (495 – 520).

The reaction is linear up to a serum creatinine concentration of 20mg/dL.

The kit is available in the following pack sizes: 2×50 & 4x50ml.
When tested on a chemistry analyzer, the reagent showed excellent calibration stability for more than 15 days.

The reagent showed excellent performance with controls. Results obtained within +/- 2SD.

Precautions and Warnings

Do not ingest or inhale. In case of contact with eyes or skin;
Rinse immediately with plenty of soap and water. In case of
severe injuries, seek medical advice immediately

Procedure

Pipette into test tubes Reagent: 500 µL / 1000 µL
Standard or Specimen: 50 µL / 100 µL

Mix, and after 30 seconds, read the absorbance A1 of the standard or specimen. After exactly 2 minutes, read absorbance A2 of the standard or the specimen.

Interference

High levels of bilirubin & haemoglobin are known to interfere with creatinine assay employing the Jaffe method.

It is recommended that the Creatinine Jaffe method should not be used for creatinine assay in haemolyzed samples from neonates,  infants, and adults—antibiotics containing cephalosporin lead to significant false-positive values.
