Principle
Fully automated systems automatically calculate the Gamma-GT activity of each sample. Results in ukat/l = Results in IU/Ix0.0167
Gamma – glutamyltransferase (GGT) transfers Y – glutamyl group of L-Y-glutamyl-3-carboxy-4-nitroanilide to glycylglycine. L-Y-glutamyl-3-carboxy-4-nitroanilide + glycylglycine.
GGT Y-glutamyl-glycyl glycine + 5-amino-2-nitrobenzoate
With AutoPure Gamma-GT, the assay is linear upto 1200 10/ (20.04 ukat/l). Determine samples with higher concentrations via the rerun function. On instruments without rerun function, manually dilute samples with higher concentrations using 0.9% NaCl or distilled/deionized water (e.g. 1+4). Multiply the result by the appropriate dilution factor (e.g.5).

Calibration
There is no significant interference in samples containing upto 30 mg/dl of bilirubin, 90 mg/dl of haemoglobin and 10 mg/dl of ascorbic acid.
The amount of 5-amino-2-nitrobenzoate liberated is proportional to the GGT activity in the specimen and is measured kinetically. Reagent Storage, Stability & Handling
For calibration, it is recommended to use C.f.a.s* or any other suitable calibrator material.
Procedure
Autopure Glucose (Hexokinase) can be used on various
automated analyzers.
Quality Control
On-Board Reagent Stability R1:21 days at 20-8°C after opening. R2:21 days at 20-8°C after opening. Protect the reagent from light and contamination.
Other Details
The kit should be stored at 2-8°C and is stable till the expiry date indicated on the label.
The reagent and standard are ready-to-use and are stable til expiry, when stored at 2- 8°C. DO NOT FREEZE THE REAGENT.
