- AutoPure Cholesterol is a ready-to-use, single liquid reagent system.
- With AutoPure Cholesterol, the assay is linear upto 1000 mg/dl (26 mmol/l}.
- AutoPure Cholesterol contains lipid clearing factor (LCF) in its formulation. The LCF clears the turbidity caused by lipemic sample and thus avoids overestimation of cholesterol.
Principle
Cholesterol in serum is available as esterified and free cholesterol.
Cholesterol esters are hydrolyzed to cholesterol and fatty acids by cholesterol esterase.
In presence of peroxidase, hydrogen peroxide oxidatively couples with 4- amino antipyrine and phenol to produce red quinonimine dye. The intensity of the red colour formed is proportional to the concentration of total cholesterol in the specimen and is measured photometrically.
Procedure
Autopure Glucose (Hexokinase) can be used on various automated analyzers.
Quality Control
Each batch of AutoPure Cholesterol 300 is assayed with at least six quality control sera-Precinorm**, Precipath**, Bio Rad I***, Bio Rad II***, Accutrol Normal and Accutrol Abnormal prior to release-, To ensure adequate quality control, it is recommended that the laboratory should use a normal and abnormal commercial reference control serum. It should be realized that the use of quality control material checks both the reagent and instrument functions together. If the control values fall outside the specified limits, each of the below criteria should be cross-checked and corrected:
• Proper instrument function – wavelength setting, light source and temperature control.
• Cleanliness of probes and cuvettes.
• Bacterial contamination of wash water used by the instrument.
A comparison of the calcium determination using AutoPure Calcium Arsenazo III and AutoZyme Calcium Arsenazo III reagent gave the following correlation (mg/dl): Linear Regression y=0.949x+0.574 r=0.941 Sy.x=0.33
Other Details
- The kit should be stored at 2-8°C and is stable till the expiry date indicated on the label.
- The reagent and standard are ready-to-use and are stable til expiry, when stored at 2- 8°C. DO NOT FREEZE THE REAGENT.
