Principle

Chloride reacts with mercuric thiocyanate to give mercuric chloride. The thiocyanate which is released reacts with ferric ions present in the reagent to form ferric thiocyanate which is directly proportional to chloride present in serum/plasma & can be measured at 510 nm.(500 – 520 nm.).

Incubation:
Incubate the assay mixture for 1 minute at room temperature
(25-30°C). After completion of incubation period measure the
absorbance of assay mixture against blank at 510 nm. The final
colour is stable for one hour if not exposed to direct light.

Quality Control

To ensure adequate quality control, it is recommended that each assay should include a normal and an abnormal commercial reference control serum. It should be realized that the use of quality control material checks both, instrument and reagent functions together. Factors which might affect the performance of this test include proper instrument function, cleanliness of glassware and accuracy of pipetting.

Other Details

The specimen to reagent ratio can be altered proportionally without affecting the results.
